Tricarboxylic Acid Cycle in Pseudomonas Aeruginosa”

چکیده

During the past few years scattered information has appeared which indicates that at least some bacteria probably possess a tricarboxylic acid cycle. Lockwood and Stodola found that Pseudomonas jluorescens formed considerable quantities of a-ketoglutaric acid when growing on a glucose medium under conditions of intense aeration (1). Ajl and Werkman found evidence for at least parts of the conventional cycle in Escherichia co2i (2). However, Karlsson and Barker (3) were not able to demonstrate the presence of this system in Azotobacter agile, either by the technique of simultaneous adaptation or by radioactive tracers. In contradiction to this latter work Stern and Ochoa (4) were able to show that both A. agile and E. wli possess the enzyme necessary for the condensation of oxalacetate and “active acetate” to citrate. Since Karlsson and Barker did not report on the activity of the organism towards the Ce members of the tricarboxylic acid cycle, it is not possible to decide whether the technique of simultaneous adaptation is applicable to the detection of the intermediates of the tricarboxylic acid cycle or whether A. agile possesses only fragments of the cycle. The present study was undertaken in an effort to detect a tricarboxylic acid cycle in Pseudomonas aeruginosa and thereby to elucidate further the pathway of glucose oxidation in this organism.

Identification of a multienzyme complex of the tricarboxylic acid cycle enzymes containing citrate synthase isoenzymes from Pseudomonas aeruginosa.

A multienzyme complex of tricarboxylic acid cycle enzymes, catalysing the consecutive reactions from fumarate to 2-oxoglutarate, has been identified in extracts of Pseudomonas aeruginosa prepared by gentle osmotic lysis of the cells. The individual enzyme activities of fumarase, malate dehydrogenase, citrate synthase, aconitase and isocitrate dehydrogenase can be used to reconstitute the comple...

Isolation and Characterization of Catabolite Repression Control

Independently controlled, inducible, catabolic genes in Pseudomonas aeruginosa are subject to strong catabolite repression control by intermediates of the tricarboxylic acid cycle. Mutants which exhibited a pleiotropic loss of catabolite repression control of multiple pathways were isolated. The mutations mapped in the li-min region of the P. aeruginosa chromosome near argB and pyrE and were de...

The Tricarboxylic Acid Cycle in Rhodospirillum Rubrum

The oxidation of acetic acid in the dark by the non-sulfur purple bacterium, Rhodospirillum rubrum, was shown by van Niel (1, 2) to require the presence of either carbon dioxide or trace amounts of the Cb-dicarboxylic acids of the Krebs cycle. These results could readily be interpreted to mean that the oxidation of acetic acid proceeds by way of the Krebs cycle, the presence of COZ being requir...

Evidence for the occurrence of Permeases for tricarboxylic acid cycle intermediates in Pseudomonas aeruginosa.

Washed suspensions and cell-free extracts of Pseudomonas aeruginosa grown on Lemco agar, to which intermediates of the tricarboxylic acid cycle had been added, were tested for their ability to oxidize succinate, fumarate, malate, pyruvate, acetate, a-oxoglutarate and citrate. Whole organisms had lag periods of 2-3 hr. before citrate was oxidized rapidly, except when citrate or acetate had been ...
